While biorelevant media are commonly used in dissolution testing, applying them to simultaneous dissolution–permeation (flux) assays bring unique challenges. In this talk, Dr. Borbás will discuss the use of human and canine biorelevant media and their impact on:

- Membrane integrity in the presence of bile micelles
- Differences in bile micelle size between species
- Effects of gastric pH and bile micelle concentration on micelle size and membrane stability

We recently teamed up with Syngenta Shradha Singh Xinghao Zhou, MRSB to demo how our eProtein Discovery™ system could help them better express difficult proteins from many sources - plant, fungi, insect, and bacteria! Using automated digital microfluidics, the team could screen dozens of proteins with different conditions to find the best one that works in just 24 hours. They even had one protein that they couldn’t express in cell-based systems that worked in our cell-free expression system.

FabRICATOR® in a correlative N-glycan analysis of cetuximab produced in different expression systems!

A strategy for comparative characterization of cetuximab produced in a variety of expression cell lines is described in this paper from scientists at the National Institute for Bioprocessing Research and Training (NIBRT) in Dublin, Ireland. FabRICATOR (IdeS) is a critical tool in this study, allowing for site-specific glycan analysis to be performed following antibody digestion into F(ab’)2 and Fc/2 subunits!
