NIPT (Non-Invasive Prenatal Test) एक Advanced Genetic Screening Test है।
यह टेस्ट माँ के रक्त में मौजूद Cell Free Fetal DNA (cffDNA) पर आधारित होता है।
यह भ्रूण के लिए पूर्णतः सुरक्षित (Non-Invasive) है।

2. टेस्ट का सिद्धांत (Principle)
गर्भावस्था के 9वें सप्ताह के बाद प्लेसेंटा से भ्रूण का DNA माँ के रक्त में प्रवेश करता है।
इस DNA का विश्लेषण Next Generation Sequencing (NGS) तकनीक द्वारा किया जाता है।
विभिन्न क्रोमोसोम से प्राप्त DNA fragments की संख्या की तुलना की जाती है।

3. स्क्रीन किए जाने वाले क्रोमोसोमल विकार
Trisomy 21 (Down Syndrome)
Trisomy 18 (Edwards Syndrome)
Trisomy 13 (Patau Syndrome)
S*x Chromosome Aneuploidy (XO, XXY, ###, XYY)

4. सैंपल की आवश्यकता
10 ml माँ का Venous Blood
EDTA या cfDNA Stabilization Tube में संग्रह
Hemolysis रहित सैंपल अनिवार्य

5. फिटल फ्रैक्शन (Fetal Fraction)
माँ के रक्त में भ्रूण DNA का प्रतिशत Fetal Fraction कहलाता है।
न्यूनतम आवश्यक मात्रा: ≥4%
कम Fetal Fraction होने पर रिपोर्ट No Call / Inconclusive हो सकती है।

6. टेस्ट की सेंसिटिविटी एवं स्पेसिफिसिटी
Trisomy 21 के लिए Sensitivity >99%
Trisomy 18 के लिए लगभग 97–99%
Trisomy 13 के लिए लगभग 90–95%
False Positive की संभावना बहुत कम

7. पॉजिटिव रिज़ल्ट का अर्थ
पॉजिटिव NIPT भ्रूण में क्रोमोसोमल विकार की संभावना दर्शाता है।
यह डायग्नोस्टिक टेस्ट नहीं है।
पुष्टि हेतु Amniocentesis या CVS आवश्यक है।

8. NIPT की सीमाएं
यह अंतिम पुष्टि नहीं करता।
Mosaicism के कारण रिज़ल्ट प्रभावित हो सकता है।
Multiple Pregnancy में सीमित उपयोगिता।
सभी Rare Genetic Disorders का पता नहीं चलता।

9. किन गर्भवती महिलाओं को NIPT की सलाह दी जाती है
माँ की उम्र 35 वर्ष या अधिक
