*SHINE YOUR LIGHT*

How do you feel when you don't get the reward, recognition or credit for your hard work? Bad?

I feel that way regularly as a medical laboratory professional. I and millions of others like me work tirelessly behind the scenes to ensure accurate diagnoses, yet we receive little to no recognition for the crucial role we play. Maybe we are invisible because our work happens in the lab, away from the public eye.

But while doctors and nurses are at the forefront of patient care, the lab scientists provide the vital data needed to make informed decisions. We analyze blood samples, diagnose infections, monitor diseases, and ensure the accuracy of critical test results that guide treatments and save lives. So the irony here is striking. Because without medical lab scientists, doctors would be left guessing. Yet, in public discourse, media, and even in healthcare celebrations, we remain in the shadows.

Well, as a friend usually says, "there will always be someone who cannot see your worth, but don't let that person be you". So maybe we should stop waiting for people to put a spotlight on us. Maybe it's time we switch on our own lights on and let it shine.

TYPES OF CULTURE MEDIA BASED ON OXYGEN REQUIREMENT.

Microorganisms have different requirements for growth depending on oxygen requirements.

1. Aerobic media
In this media, it is easy to cultivate microbes, on solid media, the growth occurs by keeping the culture in the incubator. It shows the growth; of non-fastidious microorganisms.

Examples of aerobic media are- liquid media, solid media

Peptone water- 1%peptone + 0.5% Nacl +100ml water.

Nutrient agar- nutrient broth +2% agar.

2. Anaerobic media
The media cultivates anaerobic bacteria at low oxygen, reducing oxidation-reduction potential. Anaerobic media contains extra nutrients like vitamin K, hemin, and oxygen that get reduced by a physical or chemical process. The addition of glucose (1%), thioglycollate(0.1%), ascorbic acid (0.1%), cysteine (0.05%), or iron fillings added to cause the medium to reduce. The medium is boiled in a water bath to force out dissolved oxygen and packed with sterile paraffin.

Examples of Anaerobic media:

RCM (Robertson cooked meat) isolation for Clostridium sp.

Thioglycolate broth– It has sodium glycolate that maintains low oxygen.

HOW TO PREPARE CULTURE MEDIA?

-Weigh the amount of ingredients powder on weighing machine.

-Dissolve the ingredients in distilled water.

-Adjust PH of the medium if needed.

-Add agar and boiled it to dissolve.

-Pour the media into flask.

-Autoclave the media when ingredients fully dissolve.

-Sterilization is done in autoclave to prevent from contamination, at 121ºC for 15 min at 15lbs.

-After the autoclave place the media flask in laminar air flow.

-Sterilize the laminar air flow with 70% alcohol.

-A bit cools down the media and pours into sterile Petri-plates for solidification.

-Then sample is ready to spread(spreader) / streak

-(Inoculation loop) on the medium for identification or isolation of microbes.

-Sealed the Petri plates with paraffin, label them.

-Keep them inverted in incubator at 37ºC for 24hrs.

-Observe the result next day colonies formation is visible on the media.

WHAT IS CULTURE MEDIA?

The media is a source of nutrients to support the growth of the micro-organisms in-vitro. The media helps in the growth and counting of microbial cells, selection of microorganisms, and survival of microorganisms. The culture medium can be liquid or gel.

COMMON INGREDIENTS OF CULTURE MEDIA

Peptone- source of carbon and nitrogen.

Beef extract- source of amino acid, vitamins, minerals.

Yeast extract- source of vitamin, carbon, nitrogen.

Distilled water

Agar- solidifying agent.

WHAT IS A STANDARD OPERATING PROCEDURE (SOP)?

A standard operating procedure (SOP) is a document that formalizes the tasks in an organization, linking them to the people responsible for them and to the resources used in each stage. In this way, it provides the identification of the data of “who,” “what” and “when” all phases of operations are carried out.

A SOP presents this information in a broad manner and generally using more technical language, according to the terms of legislation and standard norms.

WHAT IS A WORK INSTRUCTION? (BENCH AID)

Work instructions (WIs) are elaborated and linked to the SOP. In addition to describing the activities and those responsible for them, these documents provide more details on the tasks, including aspects such as the mode and time of ex*****on. In other words, WIs show “how” activities are carried out.

Based on a user manual, work instructions should use simple language and be written using an uncomplicated step-by-step process with information that is essential to perform the task. Images, photos and tables can be used to further facilitate employee understanding.

PREPARE BENCH h AIDS FOR EACH PIECE OF EQUIPMENT INCLUDING DAILY MAINTENANCE INSTRUCTIONS

WHY:

All the SOPs on use and maintenance of equipment have now been written. These SOPs are stored in the folder Equipment SOPs. However, it is convenient to have a simplified procedure near each piece of equipment that can remind the user of the most important steps to take. These procedures are called Bench Aids.

WHAT :

Develop Bench Aids for all relevant pieces of equipment. These are stepwise procedures for routine use of the piece of equipment; they contain the procedure for operating the piece of equipment and, if necessary, the basic preventive maintenance steps and calibration procedure that needs to be carried out regularly. Procedures that need to be carried out once in a while (preventive maintenance/calibration) don't have to be included in these Bench Aids. For this the user/person responsible for the piece of equipment/Equipment Officer can use the SOP.

Highly important: make the Bench Aids part of the SOPs: include the Bench Aids as appendixes to the SOPs (hence: the SOP version number must be printed on the bench aid as well). Every time an SOP is revised, the old bench aid needs to be revised and replaced by the new one. As the version number of the SOP is shown on the bench aid, you can easily check if the bench aid is still in accordance with the SOP.

HOW & WHO
Equipment Officer:

Prepare Bench Aids based on the SOPs for operating equipment.
Make the Bench Aids part of the SOPs by including them as appendix and giving them the same version number as the SOPs.
Laminate the copies of the Bench Aids or put them in plastic document pouches to protect them from water and other deteriorating factors and place them at a visible, logical location near the piece of equipment they have been written for.
Explain to the staff members the difference between Bench Aids and SOPs. Explain that they can find the routine procedures for operating the equipment on the Bench Aids but that they have to read the SOPs and use these for more specific procedures such as maintenance or calibration.
