HbA1c (Glycated Hemoglobin) Test
1. Objective
The objective of this test was to determine the percentage of glycated hemoglobin (HbA1c) in the patient’s blood to evaluate long-term blood glucose control.

2. Principle
The test was based on the fact that glucose in the blood reacted non-enzymatically with the amino groups of hemoglobin to form glycated hemoglobin (HbA1c). The amount of HbA1c formed was directly proportional to the average blood glucose concentration over the previous 2–3 months. It was measured using methods such as ion-exchange chromatography, immunoassay, or enzymatic analysis.

3. Materials
• EDTA whole blood sample
• HbA1c reagent kit
• Pipette
• Test tubes or cuvettes
• Centrifuge
• Colorimeter or automated analyzer

4. Procedure (Analytical)
1. Whole blood was collected in an EDTA tube and mixed gently to prevent clotting.
2. Hemolysate was prepared according to the reagent kit instructions.
3. The sample was incubated with specific reagents that separated or reacted with glycated hemoglobin.
4. The absorbance or chromatographic reading was taken using an automated analyzer.
5. The percentage of HbA1c was calculated automatically or from the calibration curve.

5. Result
The test showed the percentage of HbA1c in the blood sample.
Normal range:
• Non-diabetic: 4%–5.6%
• Prediabetes: 5.7%–6.4%
• Diabetes: ≥6.5%

6. Uses
• It was used to monitor long-term glucose control in diabetic patients.
• It helped assess the effectiveness of diabetic treatment.
• It provided an indication of average blood glucose levels over the past 2–3 months.

7. Consultation
If the HbA1c value was above the normal range, the patient was advised to consult a physician or endocrinologist for treatment modification, diet adjustment, and lifestyle management.
