For this year's European Awareness Week on Alcohol-Related Harm ( ), EUROCARE (The European Alcohol Policy Alliance), EASL - The Home of Hepatology and UEG co-organized the high-level meeting "Rethinking Prevention: A Dialogue with Youth" on November 4 at the European Parliament.

Hosted by Manuela Ripa, MEP and co-hosted by MEP Vytenis Andriukaitis, the event highlighted the urgent need to address alcohol's impact on youth. It brought together youth advocates, health professionals and policymakers to discuss how prevention policies can be more effective in meeting the needs of younger generations.

MEPs emphasized that the goal is not to control drinking choices but to empower citizens with clear, evidence-based information to make informed decisions. The active involvement of youth was recognized as essential to shaping inclusive and impactful policies.

By prioritizing youth perspectives, we are moving toward more evidence-based, targeted solutions for alcohol prevention and healthier futures for all.

🧬 Refined DNA damage targeting shows promise for homologous recombination (HR)-deficient pancreatic cancer

Up to 20% of pancreatic ductal adenocarcinoma (PDAC) patients harbor mutations in HR repair genes such as ATM, BRCA1/2, or PALB2. However, the PARP inhibitor olaparib is currently approved only for a small subset of BRCA1/2-mutated cases

🐭 🧫This preclinical study compared PARP inhibitors and optimized a multi-drug DNA damage repair interference regimen (PAD) to enhance treatment efficacy in HR-deficient PDAC

🔎 Key findings
🔸 Talazoparib demonstrated stronger PARP-trapping potency and cytotoxicity than olaparib
🔸 Talazoparib-based PAD (Talazoparib + ATR inhibitor + DNA-PK inhibitor) achieved superior tumor control vs. olaparib-based PAD in vivo
🔸 The combination was effective across ATM, BRCA1/2, and PALB2-mutated models
🔸 Comparable tolerability observed in mouse studies

💡 Clinical perspective
Talazoparib-based PAD represents a refined therapeutic strategy for HR-deficient PDAC, expanding potential benefit beyond BRCA-mutated tumors and supporting future clinical evaluation
