Lipid peroxidation, the oxidative degradation of lipids, is a major consequence of oxidative stress that affects cell membranes and other lipid-containing structures. The antioxidative action of hydrogen helps to prevent lipid peroxidation, thereby preserving the integrity and functionality of cellular membranes.

This preservation is critical because it maintains membrane fluidity and permeability, which are essential for proper cell signaling and nutrient transport. Additionally, intact cellular membranes serve as barriers against harmful substances and pathogens, safeguarding the internal environment of cells.

Furthermore, lipid peroxidation can lead to the formation of reactive aldehydes, such as malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE), which are highly reactive and can modify cellular proteins and nucleic acids. By inhibiting lipid peroxidation, molecular hydrogen reduces the generation of these reactive aldehydes, thus minimizing their damaging effects on cellular components.

In conclusion, the ability of molecular hydrogen to mitigate lipid peroxidation underscores its importance in maintaining cellular function and health. By preserving membrane integrity and reducing the formation of reactive aldehydes, molecular hydrogen contributes to cellular homeostasis and may offer protection against oxidative stress-related diseases.

Molecular hydrogen can modulate the expression and activity of endogenous antioxidant enzymes, such as superoxide dismutase (SOD), catalase, and glutathione peroxidase. These enzymes are essential for detoxifying reactive species and maintaining redox balance.

SOD is crucial for the dismutation of superoxide radicals into oxygen and hydrogen peroxide, reducing the harmful effects of superoxide radicals on cellular components. Catalase then converts hydrogen peroxide, which can be harmful at high concentrations, into water and oxygen, thereby preventing potential damage from hydrogen peroxide accumulation. Glutathione peroxidase further assists in reducing hydrogen peroxide and other peroxides by using glutathione as a substrate, thus protecting the cell from oxidative damage.

By increasing the expression of these enzymes, hydrogen enhances cellular defense mechanisms against oxidative stress. Elevated levels of SOD, catalase, and glutathione peroxidase lead to more efficient detoxification processes, allowing cells to better manage and neutralize reactive oxygen species (ROS) and other free radicals. This regulation ensures a more robust and lasting antioxidant response, which is crucial for cellular longevity and health.

Furthermore, hydrogen’s modulation of these enzymes can contribute to the repair and recovery of damaged tissues. The increased activity of antioxidant enzymes supports cellular resilience, enabling cells to withstand and recover more effectively from oxidative insults. This can be particularly beneficial in conditions characterized by chronic oxidative stress, showing positive effects on the nervous system, cardiovascular system, and inflammatory processes.

Additionally, hydrogen’s regulation of antioxidant enzymes does not interfere with the necessary signaling functions of ROS in normal cellular processes, such as cell proliferation, apoptosis, and immune responses. This selective regulation ensures that while the harmful effects of excess ROS are mitigated, their beneficial roles are preserved.

In summary, hydrogen’s ability to modulate endogenous antioxidant enzymes highlights its potential as a therapeutic agent in conditions related to oxidative stress. By enhancing the body’s natural antioxidant defenses, hydrogen helps maintain redox homeostasis, supports cellular health, and protects against a wide range of diseases linked to oxidative damage.

Effects of Molecular Hydrogen-Rich Water on Physical Exercise through Oxidative Stress Mechanisms

Oxidative stress typically occurs in the body when external stimuli induce the production of reactive substances that exceed the organism's antioxidant capacity. During physical exercise, especially high-intensity exercise, the increased metabolism leads to greater production of reactive oxygen species (ROS) and reactive nitrogen species (RNS). These reactive substances can cause oxidative modifications to lipids, proteins, and DNA, resulting in cellular dysfunction and damage.

Molecular hydrogen-rich water, which acts as a significant antioxidant, plays a fundamental role in mitigating oxidative stress and its inflammatory consequences by targeting ROS and supporting the body's defense system.

By acting as an important antioxidant, molecular hydrogen-rich water can help strengthen the body's defenses against reactive oxygen species (ROS) and reduce oxidative damage, which can further alleviate performance decline caused by ROS.

By selectively targeting the most harmful reactive species, molecular hydrogen plays a crucial role in restoring redox homeostasis. This balance is essential for maintaining cellular function and overall health.

Molecular hydrogen effectively neutralizes highly reactive hydroxyl radicals and peroxynitrite, which are not adequately countered by other endogenous antioxidants. Hydroxyl radicals are extremely reactive and can damage virtually all types of biomolecules at their formation site, making them one of the most dangerous forms of reactive oxygen species. Peroxynitrite, a powerful oxidant and nitrating agent formed by the reaction of nitric oxide with superoxide, can also cause significant damage to a wide range of cellular components.

The ability of hydrogen in molecular hydrogen-rich water to specifically target and neutralize these reactive species is crucial, as it reduces the oxidative burden on cells. This protective action helps prevent oxidative damage to critical cellular components such as DNA, proteins, and lipids. DNA damage can lead to mutations, genomic instability, and potentially carcinogenesis. Protein oxidation can cause loss of enzymatic activity, structural alterations, and impairment of cellular functions. Lipid peroxidation can compromise the integrity of cellular membranes, affecting membrane fluidity and permeability, ultimately leading to cell death.

By neutralizing hydroxyl radicals and peroxynitrite, molecular hydrogen contributes to maintaining the structural and functional integrity of cellular components. This action not only protects cells from immediate oxidative damage but also supports long-term cellular health by preventing the accumulation of oxidative damage over time. Reducing oxidative stress may also mitigate the onset of various oxidative stress-related diseases, including neurodegenerative disorders, cardiovascular diseases, and chronic inflammatory conditions.

Furthermore, the selective scavenging mechanism of molecular hydrogen does not interfere with the signaling roles of other reactive oxygen and nitrogen species, which are vital for normal cellular processes. This selective neutrality ensures that hydrogen does not disrupt essential cellular functions while providing its protective effects. Therefore, the unique properties of molecular hydrogen-rich water make it an effective and safe antioxidant for maintaining cellular homeostasis and promoting overall health.

Several studies examine the potential benefits of supplementation with molecular hydrogen-rich water on sports performance, documenting improvements in endurance, strength, sprint times, lunge movements, countermovement jump height, peak power output, repeated sprints, maximum heart rate, time to exhaustion, perceived exertion rate, distance rowed in 30 seconds, average power, and 30-meter sprint time. These results suggest that molecular hydrogen-rich water could represent a valuable ergogenic aid, potentially capable of enhancing multiple aspects of athletic performance.

A randomized, double-blind, placebo-controlled crossover study was conducted on 16 male professional soccer players, aged 18.8 ± 1.2 years. The results indicated that sprint times for the 14th and 15th 15-meter sprints were significantly faster after administration of 1260 mL of molecular hydrogen-rich water (HRW) compared to the control group, which received 1260 mL of purified water, showing improvements of 3.4% (HRW: 2.57 ± 0.12 s; placebo: 2.66 ± 0.15 s) and 2.7% (HRW: 2.57 ± 0.09 s; placebo: 2.64 ± 0.13 s), respectively, during the time window from 8:30 to 11:00 a.m. Additionally, the HRW group showed a significant 1.9% improvement (HRW: 4.54 ± 0.14 s; placebo: 4.63 ± 0.17 s) in 30-meter sprint time during the final sprint.

In a randomized, double-blind, placebo-controlled crossover study, 12 men with an average age of 23.8 ± 1.9 years performed half squats and knee extensions. These results suggest that acute intermittent hydration with molecular hydrogen-rich water improves muscle function, reduces lactate response, and alleviates delayed onset muscle soreness.

One study was conducted on eight female participants (mean age ± SD: 21.5 ± 5.0 years; VO2 max 45.0 ± 2.5 mL·kg−1·min−1) and four male participants (age 18.9 ± 1.3 years; VO2 max 52.2 ± 1.7 mL·kg−1·min−1). This investigation used a randomized, double-blind, placebo-controlled crossover design. The results indicated that, compared to placebo, molecular hydrogen-rich water led to improvements in countermovement jump height (30.7 ± 5.5 cm vs. 29.8 ± 5.8 cm; p = 0.014) and reductions in blood creatine kinase activity (156 ± 63 UL−1 vs. 190 ± 64 UL−1; p = 0.043), as well as a decrease in muscle soreness (34 ± 12 mm vs. 42 ± 12 mm; p = 0.045) 12 hours after afternoon training.

In a different randomized double-blind study, 22 male amateur middle-distance runners participated. Overall, the results suggest that intake of molecular hydrogen-rich water led to significant improvements in maximum heart rate, time to exhaustion, and perceived exertion (RPE) among amateur endurance athletes.

In another investigation, 18 dragon boat athletes, including 12 males and 6 females, participated. In conclusion, short-term consumption of molecular hydrogen-rich water demonstrated effectiveness in improving strength performance in dragon boat athletes, as well as facilitating heart rate recovery to baseline levels after exercise. These results suggest that molecular hydrogen-rich water could represent a suitable hydration strategy for athletes.

Significant Study Among Trained Amateur Cyclists and Cycling Enthusiasts on Therapy with Molecular Hydrogen-Rich Water

A randomized, double-blind, placebo-controlled, crossover study was conducted with 37 volunteers. Participants were divided into two groups: untrained individuals (n = 15; age: 26.3 ± 5.9 years, weight: 69.8 ± 11.4 kg, height: 169.3 ± 7.1 cm, body fat: 24.5 ± 6.5%) and trained amateur cyclists (n = 12; age: 25.5 ± 5.5 years, weight: 70.9 ± 8.5 kg, height: 177.3 ± 6.6 cm, body fat: 17.9 ± 5.8%). Each participant received either a placebo (purified water) or molecular hydrogen-rich water (pH 7.5; hydrogen concentration: 1.9 ppm; oxidation-reduction potential: −600 mV). Performance was assessed through incremental VO2 max tests and maximal anaerobic tests following a 7-day intake period.

Results indicated that only the trained cyclists showed performance improvements in the anaerobic test after consuming hydrogen-rich water. Specifically, they demonstrated increases in peak power (from 766.2 ± 125.6 to 826.5 ± 143.4 W; d = 0.51) and mean power (from 350.0 ± 53.5 to 380.2 ± 71.3 W; d = 0.51), along with a decrease in the fatigue index (from 77.6 ± 5.8 to 75.1 ± 5.9%; d = 0.45). These findings suggest that the ergogenic effect of hydrogen-rich water is influenced by training status and that a 7-day intake regimen may be an effective strategy to enhance anaerobic performance in trained cyclists.

In a separate investigation, eight well-trained male cyclists (mean ± SD: age: 41 ± 7 years; weight: 72.3 ± 4.4 kg; height: 1.77 ± 0.04 m; maximal oxygen consumption (VO2max): 52.6 ± 4.4 mL·kg−1·min−1) were enrolled in a randomized, double-blind, placebo-controlled, crossover study. These results confidently suggest that two weeks of consuming molecular hydrogen-enriched water can effectively preserve peak power output (PPO) during repeated sprints lasting over 30 minutes.

Queste sostanze reattive possono causare modificazioni ossidative a lipidi, proteine e DNA, risultando in disfunzioni e danni cellulari.
