Developing a robust RP-HPLC method for Tirzepatide (GLP-1 agonist)
Accurate and reproducible analytical methods are critical for GLP-1 receptor agonists, where peptide complexity, higher-order structure and impurity profiling directly impact therapeutic performance.
This application note presents a step-by-step method development strategy for Tirzepatide, highlighting how systematic optimization leads to superior resolution, peak shape and robustness:
• Mobile phase screening: Neutral to weakly alkaline conditions (20 mM ammonium acetate / acetonitrile) delivered sharper peaks and better resolution compared to acidic systems.
• Column temperature: Increasing temperature from 40 °C to 60 °C significantly improved peak width and separation efficiency.
• Column chemistry: Larger pore size Bio C18 (30 nm) columns demonstrated enhanced peak shape and impurity separation versus conventional C18/C8 phases.
• Bioinert hardware advantage: Bioinert coated columns showed improved peak symmetry and higher recovery, minimizing non-specific adsorption effects.
