Mucin Clot Test !
The Mucin Clot Test is used to assess the quality and polymerization of hyaluronic acid in synovial fluid. It indirectly evaluates the viscosity of the fluid and helps differentiate inflammatory vs. non-inflammatory joint effusions.
Synovial fluid contains hyaluronic acid, which forms a clot when mixed with acetic acid or other precipitants.
• The strength and integrity of the mucin clot reflect the degree of hyaluronic acid degradation.
• Inflammation breaks down hyaluronic acid → poor clot.

🧪 Interpretation
1️⃣ Good / Firm Mucin Clot
- Hyaluronic acid well preserved
• High viscosity fluid
• Suggests non-inflammatory conditions
• Examples: Osteoarthritis, traumatic effusion
2️⃣ Poor / Friable Mucin Clot
• Hyaluronic acid partially degraded
• Low viscosity
• Suggests inflammatory conditions
• Examples: Rheumatoid arthritis, gout, pseudogout
3️⃣ No Clot
• Severe hyaluronic acid breakdown
• Marked inflammation or infection
• Examples: Septic arthritis, purulent fluids, very high WBC count.
