Cucurbitacins, a group of the mightiest anticancer compounds

Cucurbitacins are a group of steroid like triterpenes found in the members of Cucurbitaceae like Momordica Bryonia, Cucumis, Cucurbita, Luffa, Echinocystis, Lagenaria and Citrullus and a few members of Brassicaceae (seeds of Iberis and Lepidium sativum), Scrophulariaceae, Begoniaceae, Primulaceae, Liliaceae, Tropaeolaceae and Rosaceae. The diversity of cucurbitacins lies in variety of its side chain derivatives that contribute to their disparate pharmacological actions. They may be concentrated in fruits and roots of mature plants.
Structure
All cucurbitacins contain a basic 19-(10→9β)–abeo--10α–lanost–5--ene ring skeleton characterised by a double bond at 5, a methyl group at C-9 (rather than C-10) and numerous keto--, hydroxyl--, and acetoxy--groups. Most of the Cucurbitacins are tetracyclic, but some representatives have an extra ring due to formal cyclization between C--16 and C--24 as in cucurbitacins S and T. Certain Cucurbitacins have been discovered in the form of glycosides at C2 and some of them lack C--11 carbonyl function. Chemically, Cucurbitacins are ranked according to presence of various functional groups on rings A and C, diversity in side chain and stereochemical considerations. The structural composition of following Cucurbitacins are known and have been designated by the letters: A, B, C, D, E, F, G, H, I, J, K, L, O, P, Q, R and S. Momordica contain a special group of Cucurbitacins called momordicosides differing in possessing an aldehyde at C19 The sugar linkage is generally present at C-2 (2-O-β-glycosides) in Cucurbitacin glycosides.
Curative properties
Cucurbitacins have been proved to possess curative action against many types of cancer, inflammation, artherosclerosis and diabetes.
Anticancer properies
Cucurbitacins exert anticancer effects on many types of cancers like leukemia, breast cancer, lung cancer etc through a variety of mechanisms, including inhibiting cancer cell proliferation, promoting cell cycle arrest and apoptosis, inhibiting the invasion and metastasis, and destroying the protein skeleton The most significant mechanisms with regard to the apoptotic effects of cucurbitacins are their ability to modify mitochondrial trans-membrane potential and transcriptional activities via nuclear factors or genes and their capability to activate or inhibit pro- or anti-apoptotic proteins. In general, cucurbitacins are considered to be selective inhibitors of the JAK/STAT pathways; however, other mechanisms may be implicated in their apoptotic effects, including the MAPK pathway (known to be important for cancer cell proliferation and survival), PARP cleavage, expression of active caspase-3, decreased pSTAT3 and JAK3 levels, as well as decreases in various downstream STAT3 targets such as Mcl-1, Bcl-2, Bcl-xL, and cyclin D3, all of which are implicated in apoptosis and the cell cycle control.
Anti-inflammatory activities.
Cucurcitacin analogues viz. Cucurbitacin R and DHCB have been reported to possess anti-inflammatory potential and their action is reported to be mediated by inhibition of tumor necrosis factors (TNF)-α and other mediators of inflammation such as nitric-oxide synthase-2 and cyclo-oxygenase-2.
Anti plaque activities.
There have been reports on Cucurbitacin B and E in glycosidic form to exhibit inhibitory effect on lipid oxidation products like- malonaldehyde (MDA) and 4-hydroxynonenal (4-HNE. These reports bolster the therapeutic role of Cucurbitacins in artherosclerosis, which involves modification of lipoproteins by involvement of- MDA and 4-HNE.
Anti-diabetic properties
There have been a plethora of reports on the role of Cucurbitacins for their cytotoxic, hepatoprotective, cardiovascular, and antidiabetic effects. Cucurbitane triterpenoids present in momordica fruits are noted for antidiabetic and anticancer activities, this may provide leads as a class of therapeutics for diabetes and obesity. The 5’-adenosine monophosphate-activated protein kinase (AMPK) pathway is suggested as a probable mechanism for the stimulation of GLUT4 translocation by triterpenoids from M. charantia. It is particularly interesting in relation to diabetes and obesity because activation of AMPK increases fatty acid oxidation, inhibits lipid synthesis, and can improve insulin action. An analogue of 23,24-dihydrocucurbitacin F from Hintonia latiflora has been reported to possess significant hypoglycemic and antihyperglycemic effects. The probable mechanism underlying-- antihyperglycemic effect could be stimulation of insulin release and regulation of hepatic glycogen metabolism.
Other activities
Othe beneficial properties of Cucurbitacins include Spider mite resistance, antioxidant, increasing the rat capillary permeability and to demonstrate antifertility effects in female mice, act as allomones in many plant species and anti-feedants for few insects, birds and as kairomones
Toxicity
Cucurbitacins are highly toxic compounds and instances of severe poisoning and death in sheep and cattle that consumed bitter fruits of Cucumis and Cucurbita are well documented. The range of toxicity of Cucurbitacins based on few in-vivo toxicity reports, has been found to be between 2 -12.5 mg/kg.

I am very happy to share the wonderful results of using our supplement for diabetes "Terogena' with "Manjistha", the elixir. A person having fasting glucose levels of 282 mg/dl (and PPBG of 382 mg) on 7th March (report attached), who used our Terogena and Manjishta at prescribed doses, when tested after 11 days (on 18th March), showed fasting blood glucose of 127.7 mg/dl (report attached). Terogena has been believed by many hundreds of people, but the catalysing effect of Manjishta is proved now. Though many people used this combination and recovered in the past, nobody offered me data on their test results. Once the sugar levels reach appreciable levels, the doses will be modified. Better ways to prepare effective preparations of anticancer and biopesticidal Graveola ( Annona muricata)

Annona muricata (Laxman Phal/ Hanuman phal, Mullatha in Mal) is a sought after plant known for its anticancer and biopesticidal potentials. Many patients collect leaves (found throughout year), boil them in water and drink. When fruits are available (Feb-April) they are eaten raw. Boiling leaves destroys the heat sensitive active compounds (acetogenins) and thus this extract may not give desired results at all (See properties of acetogenins below).
The anticancer activities are due to Acetogenins (also known as THF- tetrahydrofuran acetogenins) which are characterized by a hydrocarbon chain of 35-37 carbons having a -lactone ring on one end and one/two /three tetrahydrofuran rings in the middle (for e.g.. annomuricin, annonacin etc.) Two hundred and twelve bioactive compounds have been reported from A. muricata of which more than 120 are different acetogenins, 30 are alkaloids like annonaine & annomuricine and 35 are phenolics including gallic acid, apigenin and tannins. Leaves contain the maximum acetogenins (43) followed by seeds (35) and pulp (13).
Several studies reported that A. muricata extracts (mainly from leaves and seeds) have an anti-tumor effect on several cell lines of breast, prostate, liver, and lung cancer among others, with the anti-tumor activity attributed to acetogenins. Isolated extracts up-regulate the activity of caspase 3 and 8 (apoptosis effectors), while downregulating the expression of survivin and Bcl-2, thereby enhancing apoptosis. The acetogenin annonacin promotes apoptosis in cancer cells by activating the caspase 3 and Bax pathways (Yuan et al., 2003), while squamocin induces apoptosis through the expression of the proapoptotic genes Bax and Bad, which results in the cleavage of PARP and the enhanced activity of caspase 3 in bladder T24 cancer cells (Yuan et al., 2006). They also lead to cycle arrest, which has implications for the proliferation of tumor cells. Acetogenins regulate the cell cycle in the G1/S transition by inhibiting cyclin D1 expression in human hepatocellular carcinoma cells (Qian et al., 2015). In this context, the A. muricata extract arrests the cell cycle at the G1 phase and decreases the number of cells in the S phase in a concentration-dependent manner by reducing the expression of cyclin D1, an important regulatory protein of the cell cycle (Torres et al., 2012). They can also inhibit the mitochondrial Nicotinamide adenine dinucleotide (NADH) ubiquinone oxidase reductase (Complex I of the respiratory chain), which reduces the production of ATP.
However most of the people who administer this plant do not understand the properties of acetogenins. The acetogenins are low polarity compounds and they decompose at 60ºC and their extraction yields are low using classical organic solvent extraction method. Why leaves are green and rose flower is rose.
Once we study the pigments responsible for the colors of objects around us we may wonder why chlorophyll, the pigments responsible for the green color of leaves, absorbs in red region (630-700 nm- meaning it’s color is essentially red!!!) , appears green to us. Actually the wavelength at which Chlorophyll a absorbs (λ max of chlorophyll a is 663 nm and of chlorophyll b is 645nm) is actually red while it appears as green in color (chl.a is bluish green and chl.b is yellowish green). Similarly the rose flowers contain anthocyanins which absorb between 520 and 550 nm i.e. green region, in reality are green in color, appear as pinkish red (650-700). The carotenoids which absorbs in 460-480 nm region – essentially blue in color) is yellow in color (580-600 nm) to our eye!1
This paradox can be solved if we understand the theory of “Complimentary colors”. When a green leaf (Chlorophylls) absorbs in red region, the other 6 colors of VIBGYOR i.e. “VIBGYO- ” are reflected and these 6 colours (without red color) combine to produce a color compliment to red that is green. Similarly in a red flower with anthocyanins absorbs green colour, the other six colours (VIB-YOR) combine to produce a red color. When the yellow carotenoids absorb Blue color, the remaining colors (VI-GYOR) produce a complimentary color, yellow.
This gives an advantage to anyone who wants to estimate the pigments in a material. He has to look for the complimentary color region of the visible color and find out the absorption maximum (λ max) of that compound. To say simply, chlorophylls can be estimated at red region, anthocyanins in green region and carotenoids in blue region.

# Food as medicines
All the protein you eat will not get converted to body proteins.
The proteins which we ingest in our diet will get hydrolysed in our digestive tract, and the amino acids released will be absorbed and taken to cells of various parts of our body where new proteins are synthesized. Our body can synthesize 11 amino acids only and we must get the rest ,10, from our diet. These 10 amino acids which should be “essential” components of food are known as essential amino acids. They are so important in protein synthesis (protein makes up to 50% of dry matter of protoplasm) that in absence of any of these 10 amino acids, no new protein is synthesized and the amino acids get deaminated to carboxylic acids and used as energy sources. They are the following :
M – Methionine
I – Isoleucine
L – Leucine
L – Lysine
P – Phenyl alanine
A – Arginine
T – Tryptophan
H – Histidine.
T – Threonine
V – Valine
You can remember them by the acronym “Millpath TV”.
Non-vegetarian food like meat, eggs, and milk, are complete proteins containing all of the essential amino acids. But for strict vegetarians the easiest source is pulses. Pulse proteins are much better for maintenance and building up of body because of three reasons: 1. they contain all the 10 “essential amino acids”, 2. are easily soluble in water and thus easily digestible and 3. are not destroyed by cooking. In absence of any of these 10 amino acids, no new protein is synthesized and the amino acids get deaminated to carboxylic acids and get into respiratory stream.
2. Proteins in pulses are mostly (up to 80%) albumins (soluble in water) and globulins (soluble in salt solutions) always predominate over prolamines and glutelins ( both insoluble in water or salt solutions, but only soluble in dil. acid/alkali/70-90% alcohol). Although the ratio of globulin to albumin varies in different seeds, globulins are the major proteins. In Vicia faba, 80% of the total protein consists of globulins and albumins while glutelins form 15% and prolamines less than 5%. This ratio is closely followed in majority of pulses.

3) Unlike cereal proteins, the pulse proteins are not destroyed by cooking or in other words there is no Maillard or Browning reaction in which certain amino acids like lysine in proteins react with carbohydrates (starch)to form insoluble (indigestible) complexes.
Another source of essential amino acids are the leaves of certain plants like spinach (Spinacia oleracea-2.9%), cauliflower (1.9%), Asparagus shoots (young- 2.2%) and broccoli (2.8%). It is to be remembered that all leaves contain useful proteins, albeit, in small amounts.
In contrast to this, cereal proteins contain only 7-8 essential amino acids resulting in reduction in protein synthesis. The gluten which forms 70-80% of storage proteins in Barley, corn and wheat is a combined aggregate of prolamines and glutelins in the ratio of 1:1. The remainder of seed proteins in these grains consists of 50% albumins and globulins. In rice, the major group of protein is glutelins (50%) and the rest consists of equal quantities of albumins, globulins and prolamines. Oats is the only exceptional cereal in that the major protein is globulin and contain all essential amino acids. Cereal proteins undergo Maillard or Browning reaction in which amino acids like lysine (with an extra amino group) in proteins react with carbohydrates (starch) to form insoluble (indigestible) complexes making it unavailable for our body.

The Beet root colours, excellent for foods and medicines

The colours of beet root, Betacyanins are the second group of red pigments responsible for the red/purple colour of beetroot and flowers of Bougainvillea, cockscomb and Cactus. In contrast the other larger group of anthocyanins which are polyphenols, betacyanins are a group of nitrogenous compounds (like alkaloids) responsible for the red and yellow color of plants belonging to the Caryophyllidae which contain about 11,00 plants (4% of total Angiosperms). It is an enigma that they are mutually exclusive to anthocyanins in that they are never seen in plants containing anthocyanins and vice-versa. They possess an indole-2- carboxylic acid / proline N-linked to a pyridine dicarboxylic acid group through an acetyl group. Where indolic moiety is there, the compounds are red in color and are designated betacyanins. The second group where proline is present, the pigments are yellow in color and known as betaxanthins. Both betacyanins and anthocyanins absorb light between 520-550 mm.
Similar to anthocyanins, betacyanins also are glycosides. More than 50 betacyanins are reported and almost all of them are glycosides of betanidin or isobetanidin. At times, even phenolic acids (like p-coumaric acid or ferulic acid) also are found esterified to betacyanins (similar to anthocyanins). But in contrast with the anthocyanins, betacyanins are unstable in acid medium and give yellow color with alkalies. (Anthocyanin salts are stable and they give blue color with bases). E. g. Amarantin (Amaranthus), Bougainvilleins, 1-XVI (Bougainvillea) and Gomphrenins 1-VIII (Gomphrena). Betaxanthins are few in number. e. g. Indicaxanthin (Opuntia), Miraxanthins 1-VI (Mirabilis) etc.
Betacyanins are as antioxidant colors for foods and pharmaceutical products since ancient times. They are successfully used in meat substitutes, summer sausage, gelatin disserts and diary products. Beet root juice is used for intensifying the color of tomato products like tomato juice, puree and catsup. It is also found to protect liver from other hepatotoxic chemicals. Betacyanin gives best color at pH 6.3-6.6.
Betacyanins have a number of health benefits
1.Anticancer property especially against leukemia, breast cancer and prostate cancer.
2. Good for brain in that they protect neurons (neuroprotective)
3. One of the best antioxidants, better that ascorbic acid. Therefore useful for all chronic diseases like diabetes, heart problems, hypertension etc
4. Protect the lipid profile by increasing HDL and protecting the membrane lipids.
5. Protect liver from toxic chemical pollutants.
5. Aid digestion
6. Prevent Lung and skin Cancer.
