International Institute for Integrative Sleep Medicine - IIIS

World-class institute for sleep medicine, aiming to solve the mechanism of sleep/wakefulness by conducting basic to clinical research.

We spend nearly one-third of our lives asleep. The mechanism and function of sleep, however, remains unclear. Many factors such as mental illnesses, food, drugs, and emotions, can affect sleep/wake regulation. Sleep disorders are not only by themselves a major problem in modern society, but also an established risk factor for metabolic syndrome and other lifestyle diseases. Deficiencies in healthy sleep cause significant social losses, and are linked to traffic accidents due to excessive sleepiness, increased prevalence of mood disorders, increased su***de deaths, and an increased care-giving burden due to wandering and delirium in the elderly. Thus, while sleep has been a perpetual topic of scientific inquiry that keeps attracting many great minds, it is also an important field where society demands the development of strategies to remedy sleep disorders and associated diseases. We gather globally prominent scientists from multiple research fields contributing to the neurobiology of sleep. They cooperate together to elucidate the fundamental principles of sleep/wake regulation, and develop new strategies to assess and treat sleep diseases as well as the closely associated metabolic and mental disorders.

【News】11月15日（土）つくば国際会議場にて第14回世界トップレベル研究拠点プログラム（WPI）サイエンスシンポジウム「未来を変える科学の眼」が開催されました。
本シンポジウムでは、WPI全拠点が一堂に会したブース展示のほか、高校生とWPIの研究者たちによるポスター発表や、若手研究者によるサイエンストークが行われ、ポスター発表の高校生４９名を含む２４４名が参加しました。
IIISからは、小柳伊代特任助教（坂口研究室）が若⼿研究者によるサイエンストークに登壇し、野間紘久さん（中島研究室）がポスター発表を行いました。

【News】The 14th WPI Science Symposium -New Scientific eyes for changing the future- was held at Tsukuba, Ibaraki on Saturday November 15th.
At this symposium, high school students and WPI researchers gave poster presentations, young researchers gave lectures, and each center set up booth displays. The 14th symposium was a great success with 244 people, including 49 high school students in attendance.
From IIIS, Dr.Iyo Koyanagi (Sakaguchi Lab) presented at the Young Researchers' Science Talk, gave a lecture titled “How the Sleeping Brain Supports Memory.”　In addition, Mr. Hirohisa Noma (Nakajima Lab) gave a poster presentation titled “A New Form of Psychotherapy: The Development and Societal Dissemination of AI Counselors.”

【プレスリリース】感情と覚醒をつなぐ新たな神経経路を発見
感情と覚醒をつなぐ新しい神経経路として、大脳辺縁系の一部である分界条床核のニューロン（神経細胞）が、中脳の深中脳核の興奮性ニューロンを介して覚醒を誘発することを明らかにしました。この経路はストレスによる睡眠障害を引き起こす神経基盤の一つである可能性があります。

私たちの感情やストレスは、眠りや目覚めに大きな影響を与えます。これまで、感情を司る脳の扁桃体やその周辺構造が覚醒に関与することは知られていましたが、その具体的な神経メカニズムはよく分かっていませんでした。本研究では、脳の扁桃体の延長部にあたる分界条床核（BNST）のGABAというアミノ酸を神経伝達物質とするニューロン（GABA作動性ニューロン）が、中脳の深中脳核（DpMe）に存在する興奮性ニューロン（グルタミン酸作動性ニューロン）を介して、睡眠中のマウスを瞬時に覚醒させる神経経路を発見しました。
BNSTのGABA作動性ニューロンを光遺伝学的に刺激すると、ノンレム睡眠中のマウスが即座に覚醒し、その際DpMeニューロンの活動が急上昇することが観察されました。さらにDpMeのグルタミン酸作動性ニューロンを選択的に除去すると、この覚醒反応が著しく減弱しました。
本研究はさまざまな神経操作技術を駆使し、感情と覚醒を結ぶ新しい神経経路を詳細に解析したものであり、これらの結果は、精神的な刺激（恐怖やストレスなど）が睡眠を中断させる神経基盤を初めて明らかにするとともに、ストレス関連の不眠症や気分障害の治療標的の発見につながると期待されます。

【News】Dr. Masashi Yanagisawa Receives the WIRED INNOVATION AWARD 2025”
　　Dr. Masashi Yanagisawa, Director of the International Institute for Integrative Sleep Medicine (IIIS), University of Tsukuba, has received the “WIRED INNOVATION AWARD 2025“, presented by WIRED Japan.
The award honors individuals and projects that are driving innovation and shaping the future across science, technology, and society.

　　Dr. Yanagisawa has made major contributions to uncovering the mechanisms of sleep and wakefulness, including the discovery of orexin. He was also recognized for establishing a new interdisciplinary field, “sleep science,” by integrating neuroscience, experimental medicine, and pharmaceutical science. In addition, he also contributes to raising awareness and understanding in society of the fundamental question, ‘What is sleep?’

　　His appearance on WIRED Japan’s series “Tech Support” (August, 2023) remains one of the channel’s most viewed programs, with over 2.5 million views.

筑波大学国際統合睡眠医科学研究機構（IIIS）の柳沢正史機構長が、『WIRED Japan』による「WIRED INNOVATION AWARD 2025」を受賞しました。
本賞は、科学・テクノロジー・社会の分野で未来を切り拓く革新を称えるものです。

柳沢機構長は、オレキシンの発見をはじめ、睡眠／覚醒のメカニズム解明に多大な貢献を果たし、神経科学と実験医学、創薬科学を融合して「睡眠医科学」という新領域を創出した功績が評価されました。また、『睡眠とは何か』という根源的なテーマを社会に紹介し、理解を広める役割も担っています。

【Press Release】Hibernation-like Hypothermia Suppresses Neuroinflammation and Promotes Functional Recovery after Brain Injury

Researchers at University of Tsukuba have demonstrated that hibernation-like hypothermia induced by activation of hypothalamic Q neurons effectively suppresses neuroinflammation and promotes neuronal survival and motor function recovery in a murine model of traumatic brain injury (TBI). Unlike conventional therapeutic hypothermia, this physiological approach does not depend on external cooling, and thus may offer a novel and more controllable strategy for TBI treatment.

【プレスリリース】脳損傷治療に有効な冬眠様低体温状態を誘導する汎用的新手法を開発
マウス脳損傷モデルにおいて、視床下部にあるQニューロンという神経細胞を刺激して誘導される冬眠様低体温状態が、神経炎症を抑え神経細胞の生存と運動機能の回復を促進することを明らかにしました。外部冷却を用いない生理的低体温法として、外傷性脳損傷の新たな治療戦略になる可能性があります。

【News】筑波大学と株式会社S’UIMIN、アリナミン製薬株式会社は9月29日、昨年度より進めている「疲れと睡眠の関係、および抗疲労成分の効果」に関する共同研究プロジェクトの研究成果として、運動の仕方やその強さによって睡眠への影響が異なることが明らかになったと発表しました。

一般的には「疲れると眠くなる」と考えられていますが、一方で「疲れすぎると眠れない」というケースもあるなど、「疲れと睡眠の関係」についての科学的なメカニズムは、十分に明らかになってはいません。

そこで世界トップレベルの睡眠研究拠点である、筑波大学国際統合睡眠医科学研究機構と脳波測定デバイスを用いた睡眠測定サービスを提供するS’UIMIN社、アリナミン製薬株式会社が連携し、2024年9月より「疲れと睡眠の関係、および抗疲労成分の効果」に関する3か年計画の共同研究を進めています。

今回の研究では、ウエイトトレーニングの様な高強度のレジスタンス運動を行うと睡眠の質が良好となる一方で、ランニングなどの高強度の有酸素運動を行うと睡眠の質は低下するという傾向が示されました。高強度の有酸素運動では「疲れすぎると眠れない」現象が確認されましたが、レジスタンス運動では確認されず、睡眠の質が運動の強度や様式によって異なることが初めて明らかになりました。
