Gene Therapy Approaches for Blood Disorders👇

✅This schematic illustrates the overall process of gene therapy used to correct hematopoietic stem and progenitor cells (HSPCs) in patients with β-thalassemia (Bthal) and sickle cell disease (SCD).

✅Autologous HSPCs are first harvested from the patient. These cells contain the genetic defects responsible for abnormal red blood cell (RBC) production.

✅The collected HSPCs are then processed ex vivo, where different gene therapy strategies are applied to correct the underlying mutations.

✅Four major gene-correction strategies are highlighted:
🔴i. Gene addition by lentiviral vectors (LV), which introduces a functional gene copy.
🔴ii. Homology-directed repair (HDR), a precise gene-editing method that uses a DNA template.
🔴iii. Non-homologous end-joining (NHEJ), an alternative editing pathway used for targeted DNA disruption or correction.
🔴iv. Base editing, which enables single-nucleotide corrections without double-strand breaks.

✅After correction, the modified HSPCs are reinfused into the patient. These cells repopulate the bone marrow and generate corrected red blood cells, improving or resolving disease symptoms.
