Discovery of lunasin’s anti-mitotic property

In 1992, a poly-aspartyl rich peptide was first isolated and characterized by Japanese scientists in an effort to identify novel protease inhibitors from soybean seeds.

In 1996 at UC Berkeley, we isolated the gene for this poly-aspartyl rich peptide as a small subunit component of a 2s albumin gene that also contains a methionine-rich protein large subunit.

In the process of subcloning the gene for the poly-aspartyl rich peptide, its inadvertent expression in bacteria caused the abnormal elongation of bacterial cells without segmentation suggesting disruption of cell division.

To prove that the poly-aspartyl rich peptide can disrupt cell division in nucleated cells, I tagged the gene for this peptide with green fluorescent protein (bioluminescent protein from jelly fish) and transfected the gene construct into mice liver cancer cells.

Here are the first images taken in January 1997 on the anti-mitotic (anti-cell division) effect of the poly-aspartyl rich peptide (renamed lunasin from the Filipino word ‘lunas’ meaning cure or remedy). The microscopic images show a mice liver cancer cell unable to divide at the metaphase-anaphase stage of cell division causing cellular lysis and chromosomal fragmentation. Note the co-localization of GFP-tagged lunasin with the fragmented chromosomes.
