When leptin drops during weight loss, your brain responds by trying to bring your weight back up — one of the big drivers of yo-yo dieting.

So why don’t we have a medication that mimics leptin?

People have tried to make one. Multiple companies have created leptin agonists and taken them into clinical trials… but they didn’t work. There was no meaningful difference in weight loss, so the trials were abandoned.

Here’s the good news:
We don’t actually need a leptin medication. GLP-1 medications like semaglutide and tirzepatide send a strong enough signal to the brain that they override leptin entirely. With GLP-1s on board, leptin doesn’t matter anymore.
