MIT chemists have developed a protocol to rapidly produce protein chains up to 164 amino acids long. The flow-based technology could speed up drug development and allow scientists to design novel protein variants incorporating amino acids that don’t occur naturally in cells.

It is termed a covalent inhibitor of this viral cysteine protease which the virus uses to get in. The molecule is a beautiful example of a peptido mimetic looking like Val-Pro-Gln, constrained glutamine glutamic acid. Presumably the viral enzyme-SH attacks the nitrile.

It is a great example using some fancy amino acid analogues, nice work at Pfizer.

From a medicinal chemistry standpoint, non-catalytic inhibitors didn’t make sense, i.e., were outside the box 📦 of protein function or better effected outside the rate of main function, for transport or other metabolic turnover. However, careful thought like 👍 this could find something to pe*****te specific tumor cells, a problem in all cancers. More importantly, small molecule effectors with chiral bias adds specificity to the medicine profile, the power of chiral drugs. Better?they could inhibit or promote protein-protein interaction, cooperative association which increases function. Cool 😎, he’s screening new chiral buffers to change protein function. The challenge here will be proving effective dose, why protein effectors don’t make it (takes too much medicine). In cancer it could be a different story with chiral bias toward protein function. 🙏🏽Cravatt🙏🏽
