So why is fluorescence usually more accurate than Trypan Blue?

It comes down to the science of how the dyes interact with cells.

Trypan blue dye is excluded from entering cells unless the membrane is damaged, so it stains dead cells blue. However, it is unable to distinguish dead cells from debris.

When you’re working with sample types like primary cells, samples often include debris or cells of non-interest, like red blood cells.

For these sample types, we recommend using fluorescent dyes in conjunction with an automated cell counter.

Acridine Orange and Propidium Iodide (AO/PI) is a combination fluorescent dye that binds only to nucleated cells.

Acridine Orange permeates intact cell membranes, staining live nucleated cells so they fluoresce green.

Both Acridine Orange and Propidium Iodide stain dead nucleated cells, which fluoresce red due to a process called FRET quenching.

Non-nucleated cells and cellular debris are not stained, and therefore, they do not interfere with counts.

That means that AO/PI enables the specific identification of live / dead cells—even in the presence of large numbers of non-nucleated cells and cellular debris!

How does a UV-Vis Spectrophotometer work?

UV-Vis spectrophotometry is a powerful analytical technique used in various scientific fields to measure light absorbance across the electromagnetic spectrum’s ultraviolet (UV) and visible (Vis) ranges.

By measuring the intensity of light passing through a sample solution and comparing it to the intensity of the incident light, a UV-Vis spectrophotometer provides valuable information about the properties of materials and their interactions with light.

When incident light hits an object, it can be absorbed, reflected, or transmitted.

The spectrophotometer measures the intensity of light absorbed across the UV and Vis ranges.

Light transmitted through the sample is measured and compared to a reference measurement of the incident light source.

By applying the Beer-Lambert Law, which states that the amount of light absorbed is directly proportional to the concentration of the sample and the path length, the spectrophotometer can determine the concentration of specific analytes in the sample.
