Most wheat immune testing evaluates raw wheat proteins. However, wheat is rarely consumed in that form.

In this research, antibody responses were assessed not only to wheat flour, but to processed wheat products including bread, dough, and pasta. A consistent finding emerged: a meaningful subset of individuals who tested negative to raw wheat produced IgG and IgA antibodies once wheat had been baked, fermented, or cooked.

Processing alters wheat protein structure, creating new epitopes that may be more immunogenic for certain individuals. When testing does not account for these changes, clinically relevant reactions can be missed.

Most conversations around food sensitivity testing focus on which antibodies are measured.

But accuracy starts earlier, with how food proteins are prepared before testing even begins.

Methodology research has shown that traditional extraction techniques may recover only a fraction of certain processed food proteins, while improved approaches can capture far more of the antigen profile. One study developed a universal extraction procedure capable of recovering gliadins from both heat-processed and unprocessed foods, achieving approximately 95.5% recovery compared with much lower recovery using conventional ethanol methods. If relevant proteins are not successfully isolated, they never reach the testing phase and cannot be measured.

This is why Cyrex places significant emphasis on how antigens are prepared and validated in the lab. Our process includes:
- Proprietary antigen purification systems
- Optimized protein concentrations for each antigen
- Antigen-specific validation for every food target

These steps are designed to help capture a broader and more accurate representation of the proteins the immune system may actually encounter in the real world.
